Working Principle of Fermenters & Bioreactors

A fermenter works by creating and maintaining an optimized, controlled environment inside a sealed vessel where microorganisms or cells can grow and carry out the desired biological reaction. The process begins with sterilization of the fermenter vessel and all growth media to eliminate any unwanted microbial contamination. Once sterility is confirmed, the selected microorganism or cell culture is inoculated into the vessel.

During the fermentation cycle, the following parameters are continuously monitored and automatically regulated by the control system:

  • Temperature is maintained within the optimal range for the specific organism using a jacketed vessel through which heating or cooling fluid is circulated.
  • pH is monitored by in-line sensors and corrected automatically through the addition of acid or alkali dosing solutions to keep the culture in its productive range.
  • Dissolved Oxygen (DO) levels are controlled by adjusting the agitation speed and the rate of air or oxygen sparging into the vessel, ensuring aerobic organisms receive sufficient oxygen for growth and product formation.
  • Agitation is provided by a motor-driven impeller system that ensures uniform mixing of nutrients, gases, and microbial cells throughout the vessel, preventing concentration gradients.
  • Nutrient Feeding is managed through dedicated inlet ports, allowing continuous or intermittent addition of carbon sources, nitrogen sources, or other substrates as the culture demands.
  • Foam Control is handled by antifoam addition systems triggered by foam sensors, preventing overflow and contamination.
  • Exhaust Gas Management ensures that CO₂ and other metabolic gases produced during fermentation are safely vented through sterile exhaust filters.

At the end of the fermentation cycle, the broth containing the desired biological product is harvested through the outlet port and transferred to downstream processing for purification and formulation. The vessel then undergoes CIP (Clean-In-Place) and SIP (Sterilize-In-Place) procedures before the next batch begins.
